FAT for binary prep. HPLC-System

Recently YMC has assembled two systems for the purification of interferon at their facilities in Dinslaken. The customer took the opportunity to inspect and test the systems in the factory before shipping them to their production site (FAT = Factory Acceptance Test).

The two identical Kronlab systems are intended for the purification of interferon using a RPC-column with a binary gradient. Each system comprises a binary high pressure gradient pump (2000 ml/min at 125 bars) with a solenoid valve to apply the sample via the system pump. They have a module for pressure data acquisition and an air-bubble detection device on the sample line. A 4 way 2 position valve allows the column to be bypassed. A multiple wavelength UV detector with DAD features ensures the detection of the target proteins. The easily programmable fraction collector allows the use of different rack types for tubes or glass bottles. The RPC-column (500 x 150 mm ID) and its guard column (100 x 100 mm ID) are mounted as shown in the system using custom designed stainless steel racks.

During their visit to Germany, the customer took the opportunity to make some small changes to the system design and layout, to get first hand information about the products and work of YMC Europe GmbH and to gain additional training on the software and system.

Finally Kronlab provided the customer with detailed documentation of testing as well as qualification or support for IQ/OQ/PQ verification. Individual, client-specific testing and acceptance criteria were incorporated on request.
