GDP-Assay-Schematic

GDP-Assay-Schematic

The Transcreener GDP GTPase assay is a homogenous, fluorescent immunoassay that monitors the activity of GTPases along with GEF and GAP partners. The assay directly measures the amount of GDP produced in an in vitro GTPase reaction.

BellBrook Labs' Transcreener GDP GTPase assays are available with fluorescence polarization (FP), fluorescence intensity (FI), and far-red, time-resolved Forster-resonance-energy-transfer (TR-FRET) readouts that provide a safe, HTS-compatible alternative to cumbersome radioassay methods and are more sensitive and less subject to interference than colorimetric phosphate detection methods.

Based on BellBrook Labs' proprietary nucleotide immunodetection technology, the Transcreener® GDP GTPase Assays are compatible with any enzyme class that produces GDP, including monomeric small G proteins, such as CDC42, and Gα subunits of heterotrimeric G proteins, such as Gαi1.
GDP displaces tracer from a highly selective monoclonal antibody resulting in a decrease in fluorescence polarization or an increase in fluorescence intensity.

• Measure guanine nucleotide exchange factor (GEF) activity
• Measure GTPase-activating proteins (GAP) activity
• Homogenous, simple, mix-and-read assay
• Tunable dynamic range to match the target of interest
• Detect any GDP producing enzyme: GTPases, fucosyltransferases, mannosyltransferases

Three Readouts Available

• Fluorescence polarization (FP)
• Fluorescence intensity (FI)
• Time-resolved Forster-resonance-energy-transfer (TR-FRET)

Assay Features

• Easy-to-Use, Ultra-Sensitive GTPase Activity Assay
• HTS Compatible with 96, 384, and 1536-well plates
• Direct detection of GDP to monitor GTPase, GAP, and GEF Activity
• Stable assay reagents – minimum 8-hour reagent and signal stability

Far Red FP, FI &TR-FRET Readouts Validated on Major Multimode Readers

FerroFarRed - An Ion Indicator for the Specific Detection of Fe2+
Intracellular Fe2+ can react with hydrogen peroxide, thereby producing the hydroxyl radical, a reactive oxygen species (ROS). Excessive ROS causes oxidative stress, resulting in cancer and neurodegenerative diseases. FerroFarRed (also known as SiRhoNox-1 or ER-SiRhoNox) is a fluorescent probe, manufactured by Goryo Chemical, Japan, distributed in Europe by MoBiTec GmbH, Germany. FerroFarRed specifically detects labile iron (II) ions (Fe2+) and is known to react with Fe2+ only, separately from other metal

IATA's CEIV-Pharma Certification or HSA's GDP Certification?
Players throughout the pharmaceutical supply chain are experiencing growing pressure to gain accreditation for handling the high-value, temperature-sensitive cargo, as having a certification becomes more commonplace, rather than an exceptional feat in the industry. There are currently 2 different measures of standards, one is IATA's CEIV-Pharma Certification and the other is HSA's GDP Certification. There are major players on both sides of the fence which hold – GDP-compliant certificates and CEIV-pharma certifications.
Exploring the recent PQG / ECA guide to GDP
The Pharmaceutical Quality Group (PQG) issued a monograph on Pharmaceutical Distribution in 1997 and initiated planning to revise this in line with the new EU GDP regulatory guideline (a final revised version was issued in March 2013 with an effective date of 8 September 2013). Although the 2013 EU-GDP guidelines provide far more details than previous guidelines, it is clear that there was and is a need for further guidance on
